Shopify's own help pages cover the account side. They state that "By default, your online store was given a myshopify.com URL when you signed up for Shopify" (Shopify Help Center, Domains). So a new store has a working address before any custom domain is attached, and a taken .com only affects the address customers type to reach you.

Does a taken .com mean I cannot use the name on Shopify?
This page cannot answer that, because it checks domains and nothing inside Shopify. Shopify's help center is the source for what its admin accepts as a store name.
